Abstract: | Os fungos s o ub quos no ambiente e fazem parte da flora humana. Muitos
fungos do g nero Candida s o patog nicos oportunistas e causam infec es
em indiv duos imunodeprimidos ou com altera es do sistema imunit rio. A
gravidade da infec o determinada pela virul ncia do agente e pela sua
capacidade de escapar s defesas imunit rias do hospedeiro.
O incremento das infec es f ngicas nas ltimas d cadas levou a um
aumento do interesse no estudo dos fungos. A candid ase a infec o f ngica
mais frequente no homem sendo causada por fungos do g nero Candida.
A Candida albicans um fungo polimorfico podendo apresentar-se sob
a forma de levedura ou filamentosa com uma parede celular altamente
organizada. Os factores de virul ncia mais importantes s o: a produ o de
enzimas, a capacidade de ades o aos epit lios, a produ o de biofilmes e a
resist ncia fagocitose. Pensa-se que a produ o de ectofosfatases tamb m
um factor importante na instala o da infec o f ngica.
Este trabalho teve como objectivo estudar a actividade de
ectofosfatases em isolados cl nicos de C. albicans, “in vitro”, comparando a
actividade intr nseca das ectofosfatases destas estirpes com o fen tipo
invasivo, de acordo com o local de infec o (local ou sist mica).
As estirpes foram seleccionadas a partir de uma colec o de estirpes
patog nicas. Foram seleccionadas 33 estirpes de origens diversas: 13 de
l quidos est reis (b lis, liquido peritoneal, pleural, abdominal, asc tico), 8 de
sangue, 4 isolados de pontas de cateter, 3 de urina, 3 de exsudados vaginais e
2 de fezes.
A actividade enzim tica foi quantificada em tr s condi es: 30 C e
37 C em meio YPD e a 37 C com o meio RPMI 1640 suplementado com 10%
de soro fetal bovino inactivado, 23,8 mM de bicarbonato de s dio e 50 mM de
glicose. A medi o da actividade enzim tica da ectofosfatase foi feita
utilizando o m todo descrito por Kiffer-Moreira e colaboradores (2007), usando
o Kit de quantifica o de fosfatases cidas (“Acid Phosphatase Assay Kit” -
Sigma).
Os valores m dios de actividade enzim tica obtidos em estirpes
isoladas de sangue foram de 0,007 U/107cel. Em estirpes isoladas de outros
locais de infec o o valor m dio foi de 0,002 U/107cel, este valor aumenta
quando temos patologias oncol gicas associadas a baixa compet ncia
imunit ria.
Conclui-se que o valor de actividade das ectofosfatases est
dependente das condi es de imunidade do hospedeiro e do local de
instala o da infec o. Os valores mais elevados foram apresentados por
estirpes isoladas a partir do sangue, em que os doentes apresentavam um
sistema imunit rio deprimido, devido patologia de origem oncol gica. Fungi are ubiquitous in the environment and are part of human flora. Many fungi of the genus Candida are opportunistic pathogens and cause infections in immunocompromised individuals or with altered immune system. The severity of infection is determined by the virulence of the agent and its ability to evade host immune defenses. The increase of fungal infections in recent decades has led to increased interest in the study of fungi. Candidiasis is the most common fungal infection in humans, being caused by fungi of the genus Candida. Candida albicans is a dimorphic fungus that can present itself in the form of yeast or filamentous with a cell wall highly organized. The most important virulence factors are: the production of enzymes, the ability to adhere to epithelia, the biofilm production and the resistance to phagocytosis. It is thought that the production of ectophosphatases is also an important factor in to setting up the fungal infection. This work aimed to study the activity of ectofosfatases in clinical isolates of C. albicans in vitro by comparing the intrinsic ectofosfatase activity of these strains with invasive phenotype, according to the site of infection (local or systemic). The 33 strains used in this study were selected from a collection of pathogenic strains: 13 from sterile fluids (bile, peritoneal fluid, pleural, abdominal and ascitic), 8 from blood, 4 from catheter tip, 3 from urine, 3 from vaginal swabs and 2 from feces. The enzyme activity was measured in three conditions: 30°C and 37ºC in the medium YPD and at 37°C with RPMI 1640 supplemented with 10% fetal bovine serum inactivated, 23.8 mM sodium bicarbonate and 50 mM glucose. Measurement of ectophosphatases activity of was made using the method described by Kiffer-Moreira et al (2007), using the Acid Phosphatase Assay Kit (Sigma). The mean values of enzymatic activity obtained in strains isolated from blood were 0.007 U/107cels. In strains isolated from other infection sites the average value is 0.002 U/107cels, this value is increased when there are extensive malignancies associated with a low immune competence. It is concluded that the value of ectophosphatases activity is dependent on the host immunity conditions and the site of infection. The highest values were obtained for strains isolated from the blood, in which patients had a depressed immune system because of their oncologic pathology. |
