Abstract: | As diatomáceas bênticas formam biofilmes à superfície dos sedimentos
intertidais estuarinos e constituem um dos grupos de produtores primários
mais importantes. Estas microalgas têm a capacidade de se deslocar
activamente por entre as partículas do sedimento, processo associado à
libertação de substâncias extracelulares poliméricas (EPS). Os EPS são
constituídos maioritariamente por hidratos de carbono e a sua produção em
ambientes aquáticos é de extrema importância ao nível ecológico, servindo de
fonte de carbono para outros organismos (p. ex. bactérias e macrofauna) ou
aumentando a resistência à erosão e a estabilidade do sedimento. Este
trabalho pretendeu estudar o efeito da iluminação, do estado de
fotoaclimatação, e da motilidade celular na produção de EPS por diatomáceas
bênticas. A produção de EPS foi estudada em suspensões de comunidades
naturais bem como em espécies crescidas em cultura. Para a investigação dos
efeitos da motilidade celular foi utilizado o inibidor Latrunculina A.
Os resultados deste estudo mostraram que em suspensões naturais a
produção de EPS leva a uma acumulação durante um período de cerca de 24
h, após o qual os níveis de EPS se mantêm relativamente constantes. Foram
igualmente observados efeitos significativos da iluminação na produção de
EPS em culturas, dependendo do estado de fotoaclimatação das células. A
utilização do inibidor Latrunculina A permitiu concluir que, embora as células
imobilizadas continuem a libertar EPS, a motilidade celular tem um papel
significativo na produção de EPS. Benthic diatoms form biofilms on the surface of intertidal sediments and have been recognized as one of the most important primary producers in these environments. These photosynthetic microalgae have the ability to move actively through the sediment, a process which is associated with the release of polymeric extracellular substances (EPS). EPS consist mainly of carbohydrates and its production in aquatic environments have been showed as highly relevant to the ecological function of the ecosystem, as they are used as a carbon source for other organisms (e.g. bacteria and macrofauna) and increase the resistance to erosion and the sediment stability. This work aimed to study the effects of irradiance, photoaclimation state and cell motility in the production of EPS by benthic diatoms. The production of EPS was studied in suspensions of natural assemblages and in the diatoms grown in culture. To investigate the effects of cell motility inhibitor Latrunculin A was used. The results of the study showed that in natural suspensions, the production of EPS leads to accumulation during a period of 24h, after which the levels of EPS remained relatively constant. There were also significant effects of irradiance on the production of EPS in cultures, this effect being dependent of the photoacclimation state of the cells. The use of inhibitor Latrunculin A allowed to concluded that, although immobilized cell continue to release EPS, cell motility plays an important role in the production of EPS in benthic diatoms. |
