Abstract: | Os polihidroxialcanoatos (PHAs) são plásticos de origem bacteriana
biodegradáveis que apresentam características semelhantes aos plásticos
sintéticos. A preocupação actual é implementar processos e estratégias que
permitam produzir, a escala industrial, PHAs a baixa custo. Nesse sentido, o
presente trabalho aborda a produção de PHAs utilizando culturas mistas
(lamas activadas), provenientes de uma estação de tratamento de águas
residuais, em reactores descontínuos sequenciais sob condições de
alimentação dinâmica aeróbia.
Ao longo do trabalho foram testadas diferentes condições de operação
e de alimentação dos reactores. Os reactores foram alimentados com ácidos
orgânicos voláteis (AOVs) provenientes da acidificação de soro de leite e
outros com mistura sintética de AOVs numa proporção de HAc:HPr 2:1.
Verificou-se uma maior estabilidade na operação dos reactores alimentados
com ácidos sintéticos. Os reactores foram operados com diferentes cargas
orgânicas, e segundo os resultados obtidos quando as cargas são mais baixas
(1g CQO/L.d e 2g CQO/L.d) apresentam condições mais favoráveis à selecção
de bactérias produtoras de PHAs e maior percentagem de remoção de CQO.
Os biorreactores operaram em condições de “fome/fartura”
(alimentação dinâmica aeróbia) com ciclos de 12 e 24h. Os resultados obtidos
indicam que quando as culturas são alimentadas em ciclos de 24h a remoção
de CQO é mais rápida. A acumulação de PHAs foi confirmada por microscopia
de epifluorescência utilizando como corante o azul de Nilo. Polyhydroxyalkanoates (PHAs) are bacterial biodegradable plastics that have similar characteristics to synthetic plastics. The implementation of processes and strategies to produce PHAs at an industrial scale at low cost is an important factor for their commercialization. In this sense, this work intended to study the production of PHAs by mixed cultures (activated sludge) from wastewater treatment plant in sequential batch reactors under aerobic dynamic feeding conditions. Throughout the work different operating and feeding conditions on reactors were tested. The reactors were fed with volatile fatty acids (VFAs) from the acidification of whey and others with a synthetic mixture of VFAs with a 2:1 HAc:HPr ratio. There was greater stability in the operation of reactors fed with synthetic acids than with the fermented stream. The reactors were operated at different organic load rates. When the load rates were lower (1 g COD/L.d and 2 g COD/L.d) a greater percentage of removal of COD was observed indicating more favorable conditions for the selection of PHAsproducing bacteria. The bioreactors were operated under “feast/famine” conditions (dynamic aerobic feeding) with 12 and 24 cycles. The results showed that when the cultures were submitted to 24 hours cycles the COD removal was faster than during the 12 hours cycles. Accumulation of PHAs was confirmed by epifluorescence microscopy using the blue Nile staining. |
