Abstract: | Biomateriais são todos os tipos de materiais capazes de interagir com
sistemas biológicos para tratar, fortalecer ou substituir um tecido, um órgão ou
uma função corporal. A necessidade de desenvolver novos biomateriais,
produtos e técnicas cirúrgicas tem vindo a crescer de forma contínua.
Recentemente, os grânulos porosos têm recebido especial atenção na área da
investigação bem como a nível industrial.
No trabalho desenvolvido na presente tese, produziram-se grânulos porosos
esféricos, através da combinação de dois métodos, nomeadamente, a
introdução de um agente porogénico e a gelatinização ionotrópica, a partir de
pós de fosfatos de cálcio dopados com magnésio e estrôncio. Estudou-se o
efeito da variação de algumas condições experimentais, tendo-se observado
que a introdução dos iões dopantes na composição dos fosfatos de cálcio
alterou as características físico-químicas dos grânulos.
O trabalho realizado incluiu também a incorporação de uma molécula
terapêutica nos grânulos porosos, a levofloxacina. Dos resultados obtidos
verificou-se que os grânulos dopados com estrôncio foram os que
apresentaram o melhor perfil de libertação.
Para a avaliação do comportamento in vitro dos materiais desenvolvidos,
realizaram-se estudos de imersão dos grânulos porosos numa solução
simuladora do plasma humano, e os resultados obtidos mostraram a formação
rápida de uma camada superficial de apatite.
Os grânulos foram testados em linhas celulares pré-osteoblásticas e foram
avaliadas a viabilidade/proliferação das células, a secreção de fosfatase
alcalina e a morfologia destas. Estes ensaios mostraram que os grânulos
induzem a maturação celular.
Considerando as características microestruturais e o desempenho in vitro dos
grânulos obtidos neste trabalho, estes podem apontar-se como materiais
promissores para aplicações em engenharia de tecidos, nomeadamente na
regeneração do tecido ósseo e na libertação controlada de fármacos. Biomaterials are all kinds of materials capable of interacting with biological systems to treat, strengthen or replace a tissue, an organ or a bodily function. The need to develop new biomaterials, surgical techniques and products has been growing steadily. Recently, porous granules have received special attention in research and at industrial level. In this thesis were produced spherical porous granules, through a combination of two methods, namely the introduction of a porogenic agent and the ionotropic gelation, from calcium phosphate powders doped with magnesium and strontium. The effect of different experimental conditions was studied and it was observed that the introduction of the doping ions into the calcium phosphates composition changed the physical and chemical characteristics of the granules. This work also included the incorporation of a therapeutic molecule in porous granules, levofloxacin. From the obtained results it can be noted that the strontium doped granules had the best release profile. To evaluate the in vitro behaviour of the developed materials, immersion studies of the porous granules in a solution that simulates the human plasma were performed, and the results showed the rapid formation of an apatitic surface layer. The granules were tested in pre-osteoblastic cell lines and the cells viability/proliferation, secretion of alkaline phosphatase and morphology were evaluated. These experiments showed that the granules induce cell maturation. Considering the microstructural characteristics and the in vitro performance of the granules obtained in this work, they can be pointed as promising materials for applications in tissue engineering, particularly in bone tissue regeneration and in controlled drug delivery. |
