Abstract: | O principal objectivo deste trabalho consistiu no estudo da influência do
tratamento hiperbárico na hidrólise enzimática de xilanas presentes na pasta
kraft de Eucalyptus globulus branqueada. A hidrólise foi efectuada utilizando
uma endo enzima (EC 3.2.1.8) proveniente do fungo Thermomyces
lanuginosus designada comercialmente por Pentopan Mono BG. Previamente
à reacção de hidrólise, a pasta celulósica foi submetida a pressões entre 200-
400 MPa durante intervalos de 15 e 45 minutos. A formação de açúcares
redutores foi quantificada ao longo do tempo de reacção pelo método de DNS.
Os resultados obtidos demonstram que o tratamento a alta pressão é
responsável por promover um aumento da intensidade de hidrólise enzimática
no intervalo de pressões entre 200 e os 400 MPa. A melhor combinação
pressão/tempo foi obtida para a pressão de 400 MPa e para 15 minutos de
pré-tratamento. Para esta pasta, nos primeiros 15 minutos de reacção,
observou-se um aumento da quantidade de açúcares redutores formados de
cerca de 850% face à pasta sem pré-tratamento hiperbárico.
A análise microscópica das fibras hidrolisadas desta amostra de pasta prépressurizada
revelou que a superfície das fibras celulósicas aparenta estar
gelificada com uma camada larga e transparente, que pode ser interpretada
como sendo um indicador de perda de integridade das microfibrilas.
Ficou provado que o pré-tratamento de pastas celulósicas com alta pressão
provoca uma maior acessibilidade das fibras face à hidrólise enzimática com
xilanase, permitindo degradar fracções de xilanas mais inacessíveis. Sendo
assim, o pré-tratamento hiperbárico pode ser considerado como um
instrumento interessante na engenharia das reacções enzimáticas de pastas
celulósicas, pois variando as pressões e os tempos de tratamento conseguese
controlar a intensidade da hidrólise enzimática.
Este trabalho mostrou que o pré-tratamento hiperbárico de pastas celulósicas
é uma alternativa viável face aos métodos de hidrólise enzimática
convencionais, pois permite superar os entraves existentes na actualidade em
termos de rendimento de degradação de xilanas, permitindo ainda o controlo
da extensão da reacção de hidrólise. The main objective of this work was to study the influence of hyperbaric treatment in the enzymatic hydrolysis of xylan present in Eucalyptus globulus kraft bleached pulp. The hydrolysis was performed using an endo enzyme (EC 3.2.1.8) from the fungus Thermomyces lanuginosus, commercially known as Pentopan Mono BG. Prior to the reaction, the cellulosic pulp was subjected to pressures between 200-400 MPa during 15 or 45 minutes. The formation of reducing sugars was quantified along the reaction time by the method of DNS. The results demonstrate that the treatment the high pressure is responsible for promoting an increase of the enzymatic hydrolysis intensity in the interval of pressures between the 200 and 400 MPa. The best combination pressure/time was obtained for the pressure of 400 MPa and 15 minutes of treatment. For this case, in the first 15 minutes of reaction, there was an increased amount of reducing sugars formed from about 850%, comparing with the pulp that was not subjected to the hyperbaric treatment. Microscopic analysis of the hydrolysed kraft pulp fibers pre-treated at 300 MPa during 45 minutes revealed that the surface of cellulosic fibers appears to be gelled with a broad and transparent layer that can be interpreted as indicator of loss of the integrity of microfibrils. It was proven that the hyperbaric pre-treatment of pulps promotes an increase in the accessibility of the fibers towards the enzymatic hydrolysis with xylanase, enabling the degradation of the more inaccessible xylan fractions. Thus, the hyperbaric treatment can be considered as a useful tool in the engineering of the enzymatic reactions of cellulosic pulps, because varying the pressure and the pressurization time of the pre-treatment it is possible to control the extension of the enzymatic hydrolysis. This study showed that the pre-hyperbaric treatment of cellulosic pulp represents a viable alternative compared to the conventional methods of enzymatic hydrolysis allowing to overcome the actual problems in terms of the limited performance of degradation of xylans. |
