Abstract: | Soil contamination is a hidden danger right below our feet that extends
far beyond what we can properly measure. In Portugal, it was
estimated that approximately 2,000 sites were polluted, however, the
data regarding risk or contamination mapping is non-existent, which
makes the real situation of the national soils unknown. Species of the
genus Aspergillus are ubiquitous saprophytic fungi that have both
positive and negative impacts on human life. This type of fungi is a
constant in several world biomes, particularly soil. Due to the limited
literature focusing on the genus Aspergillus diversity in
agroecosystems, this study aimed to tackle these gaps by focusing on
the diversity of Aspergillus species in contaminated horticultural and
agricultural soil areas.
To attain this, 473 isolates were obtained from three contaminated soil
samples and observed under a stereomicroscope for an initial
screening, leaving 107 isolates to undergo further analysis. To
examine their genetic diversity, all 107 isolates were submitted to
Microsatellite - primed Polymerase Chain Reaction fingerprinting.
Based on the cluster analysis of Microsatellite - primed Polymerase
Chain Reaction fingerprints, 33 representative isolates were chosen
for Internal Transcribed Spacer amplification and sequencing. An
initial identification was accomplished with Internal Transcribed
Spacer sequences, which allowed the identification of 7 different
Aspergillus sections, with Usti and Terrei sections being the most
common. A second complementary analysis was proceeded with
amplification and posterior sequencing of the partial b-tubulin gene.
The isolates identified as members of the genus Aspergillus were
additionally subjected to multi-locus sequence analysis to accurately
identify them at species level.
Combined Internal Transcribed Spacer and b-tubulin phylogenetic
analysis allowed to identify nine species, such as A. melleus, A.
gorakhpurensis, A. udagawae, A ardalensis, A. terreus, A. hortae, A.
calidoustus, A. pseudodeflectus and A. insuetus.
Aspergillus diversity throughout the country’s contaminated soils and
their pathogenic potential towards humans deserves further
investigation. contaminação do solo é um perigo oculto que se encontra logo debaixo dos nossos pés e que se estende muito além do que podemos medir adequadamente. Em Portugal, estimou-se que cerca de 2,000 locais estavam poluídos, no entanto, a falta de dados sobre mapeamento de risco ou contaminação é inexistente, o que torna desconhecida a situação real dos solos nacionais. Espécies do género Aspergillus são fungos saprófitos ubíquos que têm impactos positivos e negativos na vida humana. Este tipo de fungo é uma constante em vários biomas mundiais, principalmente no solo. Devido à literatura limitada com foco na diversidade do género Aspergillus em agroecossistemas, este estudo teve como objetivo abordar essas lacunas, concentrando-se na diversidade de espécies de Aspergillus em áreas de solos agrícolas e hortícolas contaminados. Para atingir isso, 473 isolados foram obtidos de três amostras de solos contaminados e, numa triagem inicial, observados ao estereomicroscópio, restando 107 isolados para uma análise subsequente. Para examinar a sua diversidade genética, todos os 107 isolados foram submetidos a Microsatellite - primed Polymerase Chain Reaction fingerprinting. Com base na análise de agrupamento executada a partir de Microsatellite - primed Polymerase Chain Reaction fingerprinting, 33 isolados representativos foram escolhidos para amplificação de Internal Transcribed Spacer e sequenciamento adicional. Foi realizada uma identificação inicial com sequências Internal Transcribed Spacer, o que permitiu a identificação de 7 secções diferentes de Aspergillus, sendo as secções Usti e Terrei as mais usuais. Procedeu-se uma segunda análise complementar com amplificação e posterior sequenciamento do gene parcial de b-tubulina. Os isolados identificados como membros do género Aspergillus foram adicionalmente submetidos à análise de sequências multilocus para identificá-los com precisão ao nível de espécie. A análise filogenética combinada de Internal Transcribed Spacer e b-tubulina permitiu identificar nove espécies, nomeadamente A. melleus, A. gorakhpurensis, A. udagawae, A ardalensis, A. terreus, A. hortae, A. calidoustus, A. pseudodeflectus, A. insuetus. A diversidade de Aspergillus nos solos contaminados do país e o seu potencial patogénico para com os seres humanos merecem uma investigação mais aprofundada. |
