Abstract: | Melanoma is the most dangerous skin cancer and the incidence is
progressively increasing. Melanoma possesses high resistance to therapies
and if it is discovered in an advanced stage, treatment options are scarce and
have low efficiency. Upconversion nanoparticles (UCNPs) attract attention in
cancer theranostics due to anti-Stokes optical property and high luminescence
stability. In this work, NaYF4:Yb,Er@mSiO2 (~ 75 nm) were successfully
functionalized with folic acid for tumor targeting and loaded with doxorubicin
hydrochloride (DOX). The drug loading and controlled release performance of
the NPs were evaluated at different pH values (pH of 7.4; 5.2; and 4.2).
NaYF4:Yb,Er@mSiO2-FA demonstrated good biocompatibility to all cell lines
demonstrating above 70% of cellular viability at concentration 200 μg/mL.
Furthermore, the in vitro cytotoxic effect of DOX-loaded UCNPs against 4
different melanoma cell lines (A375, B16-F10, MNT-1 and SK-MEL-28) was
evaluated and the impact on the cell cycle dynamics were investigated in detail.
In summary, our results revealed that the nanoparticles in the study could be
used as drug delivery systems to melanoma therapy. Melanoma é o cancro de pele mais perigoso e a sua incidência está a aumentar progressivamente. O melanoma possui elevada resistência ás terapias, e se for diagnosticado em estágio avançado, as opções de tratamento são escassas e possuem baixa eficiência. As nanopartículas de conversão ascendente (UCNPs) atraíram atenção para serem usadas como teranósticos de cancro devido à sua propriedade óptica anti-Stokes e elevada estabilidade luminescente. Neste trabalho, NaYF4:Yb,Er@mSiO2 (~ 75 nm) foram funcionalizadas com ácido fólico para reconhecer células tumorais e carregadas com cloridrato de doxorrubicina (DOX). O carregamento e libertação controlada de DOX foram avaliadas a diferentes pHs (pH of 7.4; 5.2; and 4.2). NaYF4:Yb,Er@mSiO2-FA demonstrou boa biocompatibilidade em todas as linhas celulares, demostrando acima de 70% de viabilidade celular na concentração de 200 μg/mL. Posteriormente, foi avaliado o efeito citotóxico in vitro das UCNPs carregadas com DOX em 4 linhas celulares de melanoma (A375, B16-F10, MNT-1 e SK-MEL-28) e o seu impacto na dinâmica do ciclo celular. Em resumo, os nossos resultados revelaram que as nanopartículas em estudo podem ser usadas como sistemas de libertação de fármacos para a terapia do melanoma. |
