Abstract: | A utilização de biofármacos tem aumentado ao longo dos anos. Em particular, o
interferão alfa-2b apresenta atividades antiviral, antiproliferativa e imunomodeladora,
encontrando-se indicado para o tratamento de diversas neoplasias e infeções virais. Não
obstante a importância clínica do interferão alfa-2b, o seu custo é ainda relativamente
elevado devido essencialmente à falta de estratégias downstream económicas, pelo que
se torna necessário o desenvolvimento de processos de fabrico capazes de fornecer
biofármacos com alta pureza e atividade biológica que atendam os requisitos das
agências reguladoras. Face ao exposto, o principal objetivo deste trabalho consiste na
preparação de líquidos iónicos suportados em sílica e posterior aplicação para a
extração e purificação seletivas do interferão alfa-2b a partir de extratos recombinantes
de E. coli. Desta forma, foram sintetizados líquidos iónicos suportados em partículas
de sílica esférica, nomeadamente, [Sil][C3C1Im]Cl, [Sil][N3114]Cl, [Sil][N3116]Cl,
[Sil][N3118]Cl, [Sil][N3222]Cl, [Sil][N3444]Cl, [Sil][N3666]Cl e [Sil][N3888]Cl. A
caracterização destes materiais demonstrou que os líquidos iónicos se encontravam
corretamente imobilizados no suporte, mas com diferentes graus de imobilização. Após
produção e recuperação do interferão alfa-2b a partir de corpos de inclusão, foi
delineada uma estratégia cromatográfica para a sua purificação através da otimização
da composição da fase móvel (força iónica e pH), visando explorar maioritariamente a
presença de interações iónicas ou hidrofóbicas entre a proteína alvo e os SILs
sintetizados. Após várias otimizações em batch, os resultados mais promissores foram
obtidos com o [Sil][C3C1Im]Cl , permitindo a eluição do interferão alfa-2b com uma
pureza aceitável com 0,35 M de NaCl (injeção da amostra com Tris 10 mM pH 8,
seguido de eluição com 0,35 e 1,5 M de NaCl) e com 1,0 M de sulfato de amónio
(injeção da amostra com 1,8 M de sulfato de amónio, seguida de eluição com 1,0 e 0,5
M de sulfato de amónio e Tris 10 mM), em condições que favoreciam principalmente
interações iónica e hidrofóbicas, respetivamente. As condições referidas foram
aplicadas em coluna, sendo de realçar o resultado promissor na purificação do
interferão alvo no SIL já referido, bem como ao nível da reprodutibilidade dos
resultados e regeneração do suporte. Além disso, deve ser mencionado, que outros SILs
também exibiram um desempenho de purificação melhorado, não se comportando
apenas como trocadores de aniões e ligantes hidrofóbicos, mas também exibindo um
comportamento multimodal. Em conclusão, este trabalho demonstrou o alto potencial
apresentado pelos SILs para serem aplicados como fases estacionárias na cromatografia
líquida preparativa de proteínas terapêuticas. Contudo, para fortalecer esta
possibilidade, esforços futuros devem incluir a avaliação da citotoxicidade, da
estabilidade e atividade proteica, entre outros aspetos. The use of biopharmaceuticals has increased over the years. In particular, interferon alfa-2b has antiviral, antiproliferative, and immunomodulating activities, and is indicated for the treatment of several neoplasms and viral infections. Despite the clinical importance of interferon alfa-2b, its cost is still relatively high due to the lack of cost-effective downstream strategies, being therefore necessary to develop less expensive manufacturing processes able to provide biopharmaceuticals with high purity and biological activity that meet the requirements of regulatory agencies. In view of the above, the main objective of this work is the preparation of ionic liquids supported on silica and their subsequent application for the selective extraction and purification of interferon alfa-2b from recombinant E. coli extracts. In this way, ionic liquids supported on spherical silica particles were synthesized, namely, [Sil][C3C1Im]Cl, [Sil][N3114]Cl, [Sil][N3116]Cl, [Sil][N3118]Cl, [Sil][N3222]Cl, [Sil][N3444]Cl, [Sil][N3666]Cl and [Sil][N3888]Cl. The characterization of these materials demonstrated that the ionic liquids were correctly immobilized on the support, but with different degrees of immobilization. After production and recovery of interferon alfa2b from inclusion bodies, a chromatographic strategy for its purification was outlined by optimizing the mobile phase composition (ionic strength and pH), envisaging to explore mostly the presence of ionic or hydrophobic interactions between the target protein and the synthesized materials. After several optimizations in a batch mode, the most promising results were obtained with [Sil][C3C1Im]Cl, allowing the recovery of interferon α-2b with acceptable purity with 0.35 M NaCl (sample injection with Tris 10 mM pH 8, followed by elution with 0.35 and 1.5 M NaCl) and 1.0 M ammonium sulfate (sample injection with 1.8 M ammonium sulfate, followed by elution with 1.0 and 0.5 M ammonium sulfate, and Tris 10 mM), respectively in conditions that mainly favour the establishment of ionic and hydrophobic interactions. The referred conditions were applied in column tests, highlighting the promising result for the purification of interferon alfa-2b in the referred material, as well as in terms of the reproducibility of the results and regeneration of the support. It should be additionally remarked that along with [Sil][C3C1Im]Cl, other SILs also display an improved purification performance, behaving not only as anion-exchangers and hydrophobic ligands, but also displaying a multimodal chromatographic behaviour. In conclusion, this work demonstrates the high potential displayed by SILs to be applied as stationary phases in preparative liquid chromatography of therapeutic proteins. However, to strengthen this possibility, future endeavours should comprise the assessment of cytotoxicity, stability, and protein activity, among other aspects. |
