Abstract: | As fermentações vínicas são constituídas por consórcios microbianos, com
diferentes espécies e estirpes de leveduras, onde a Saccharomyces
cerevisiae tem um papel fundamental. Assim é importante avaliar a
biodiversidade intrínseca das estirpes de S. cerevisiae, por forma a explorar
o seu potencial biotecnológico. O objetivo deste trabalho foi estudar a
biodiversidade fenotípica e metabólica de S. cerevisiae, com especial
enfoque no seu potencial enológico. Para tal, isolaram-se leveduras vínicas
endógenas de 4 regiões Demarcadas portuguesas: Alentejo, Bairrada,
Douro e Dão. Isolaram-se 1260 leveduras, das quais 883 eram S.
cerevisiae. Relativamente à caracterização fenotípica, avaliou-se a
capacidade de crescimento de 313 estirpes de S. cerevisiae em 37
condições. As condições de crescimento foram selecionadas por forma a
expor a variabilidade intrínseca de S. cerevisiae, e incluíram ainda
fenótipos relacionados com o potencial enológico, nomeadamente o
crescimento a diferentes temperaturas e na presença de etanol, a produção
de H2S, e a tolerância ao cobre e ao dióxido de enxofre. Este estudo
evidenciou a existência de 292 fenótipos distintos entre a população
estudada. A caracterização metabólica foi focada na análise de metabolitos
voláteis produzidos por um conjunto de 4 estirpes de S. cerevisiae: 2
isolados vínicos e 2 estirpes comerciais amplamente utilizadas pela
indústria. A análise foi realizada por HS-SPME-GC×GC-ToFMS, onde se
identificaram um total de 218 metabolitos, pertencentes a 15 famílias, que
são diferencialmente produzidos pelas estirpes analisadas. Quer os álcoois
quer os ésteres foram as famílias com maior número de compostos
identificados, no entanto foram os compostos terpénicos que apresentaram
maior variabilidade inter-estirpe.
A análise compreensiva da variabilidade intrinseca de S. cerevisiae agora
realizada revelou uma elevada biodiversidade. Além disso, identificou-se
um metaboloma volátil de S. cerevisiae até agora desconhecido e que
demonstra a importância da continuidade do seu estudo em maior
profundidade. Wine fermentation is dependent upon a microbial consortium, with different species and strains of yeasts, which Saccharomyces cerevisiae has a fundamental role. Thus, it is important to evaluate the intrinsic biodiversity of these yeasts to explore their biotechnological potential. The aim of this work was to study phenotypic and metabolic biodiversity of S. cerevisiae, and its oenological potential. For that, S. cerevisiae strains, from Alentejo, Bairrada, Douro and Dão Portuguese Appellations were isolated. A total of 1260 wine yeasts were isolated of which 883 belong to the S. cerevisiae specie. For the phenotypic characterization, we assessed the growth capacity of 313 strains of S. cerevisiae under 37 different conditions. These growth conditions were selected in order to expose the intrinsic variability of S. cerevisiae, and also included phenotypes related to its oenological potential, particularly the growth at different temperatures and in the presence of ethanol, the H2S production, and the tolerance to copper and sulfur dioxide. This study revealed the existence of 292 distinct phenotypes within the studied population. The metabolic characterization was focused on the analysis of volatile metabolites produced by a set of 4 strains of S. cerevisiae: 2 isolates and 2 commercial strains widely used by the industry. The analysis was performed by HSSPME- GC×GC-TOFMS, where a total of 218 metabolites, belonging to 15 families, were identified and were differentially produced by studied strains. Alcohols and esters were the families with the largest number of identified compounds; whereas the terpenic compounds were those that showed the highest inter-strain variability. Indeed, the comprehensive analysis of the intrinsic variability of S. cerevisiae has unveiled a surprisingly high biodiversity. Moreover, we have identified a yet unknown volatile metabolome of S. cerevisiae, which would be of great importance to continue studying. |
