Abstract: | A detecção de agentes infecciosos em suínos tem tido um papel cada
vez mais relevante no sucesso económico das suiniculturas. O presente
estudo teve como base a identificação molecular de 12 agentes patogénicos
responsáveis por patologias em suínos. Este trabalho teve como objectivos: a
optimização de técnicas de tipagem molecular de agentes infecciosos; a
determinação de frequências de infecção de bactérias e vírus e a sua
prevalência na população suína. A avaliação dos diferentes agentes
infecciosos foi realizada em amostras de soro de 100 animais provenientes
de diferentes suiniculturas. Para a cada um destes agentes foi desenvolvida
uma técnica de tipagem molecular específica. Todas as amostras foram
tipadas por PCR para os diferentes agentes infecciosos: Actinobacillus
pleuropneumoniae; Género Brachyspira; Género Chlamydia; Género
Leptospira; Género Salmonella; Haemophilus parasuis; Influenza A; Lawsonia
intracellularis; Mycoplasma hyopneumoniae; Porcine circovirus type 2;
Pasteurella multocida; Vírus da Síndrome Respiratório e Reprodutivo dos
Suínos. Neste estudo os vírus PRRS, Influenza A e PCV2 foram os agentes
patogénicos com maior percentagem de incidência na amostragem, na ordem
dos 33 a 35%. Por outro lado, o género Salmonella apresentou a menor taxa
de incidência amostral com apenas 1% de positividade. Os restantes agentes
infecciosos alcançaram níveis de prevalência muito variáveis entre os 5 e os
18%. Os resultados alcançados neste estudo provaram estar de acordo com
os referenciais Portugueses e Europeus, sendo não significativas as
diferenças verificadas. Verificou-se ainda, que os 12 agentes infecciosos
estudados são responsáveis por causar doenças respiratórias, reprodutivas e
intestinais em suínos, sendo as mais prevalentes as doenças respiratórias.
Concluindo, os testes de detecção molecular dos agentes infecciosos são um
bom método de screening de microrganismos em suínos, apresentando bons
níveis de sensibilidade e especificidade. The detection of infectious agents in pigs plays a progressively important role in the economic success of pig farms. This study was based on the molecular identification of 12 pathogens agents responsible for pigs diseases. This work aims were: infectious agents molecular typing optimization, bacteria and viruses infection frequencies determination and their prevalence rates. Assessment of the different infectious agents was performed on serum samples from 100 animals from different pig farms. For each of these agents a specific molecular typing technique was developed. All samples were typed by PCR for the different infectious agents: Actinobacillus pleuropneumoniae; Brachyspira sp., Chlamydia sp., Leptospira sp., Genus Salmonella, Haemophilus parasuis, Influenza A, Lawsonia intracellularis, Mycoplasma hyopneumoniae, Porcine circovirus Type 2, Pasteurella multocida; Swine Reproductive and Respiratory Syndrome Virus. In this study the PRRS, Influenza and PCV2 virus were the pathogens with the highest incidence rate in our sample, with 33 to 35% rates. On the other hand, the genus Salmonella had the lowest incidence rate sampling with only 1% positivity. The other infectious agents have achieved levels of prevalence varying widely between 5 and 18%. The results achieved in this study proved to be in agreement with the Portuguese and European benchmarks, with no significant differences observed. It was also found that the 12 studied infectious agents are responsible for causing respiratory diseases, reproductive and intestinal tracts of pigs, the most prevalent respiratory diseases. In conclusion, molecular testing for infectious agents is a good microorganisms screening method for pigs, showing good levels of sensitivity and specificity. |
