Abstract: | Neste trabalho prepararam-se e caracterizaram-se hidrogéis termosensíveis
nanocompósitos de gelatina e de k-carragenano, contendo nanopartículas de
ouro como fase dispersa e procedeu-se ao estudo da cinética de libertação de
um fármaco modelo.
Estudou-se o efeito do teor e da morfologia (nanoesferas ou nanobastonetes)
das nanopartículas de ouro nas propriedades ópticas, térmicas,
intumescimento e propriedades mecânicas dos nanocompósitos. Os
nanocompósitos apresentavam propriedades ópticas características da fase
dispersa. Nos nanocompósitos de k-carragenano, a adição de nanopartículas
resultou num aumento da temperatura de fusão do gel e num aumento do
módulo elástico, indicando que as nanopartículas actuam como reforço da
matriz polimérica. Nos nanocompósitos de gelatina observou-se o
comportamento oposto, sugerindo um efeito de inibição da reticulação da
gelatina por parte das nanopartículas. Os resultados sugerem que o
intumescimento dos nanocompósitos depende da “força” do gel e do efeito de
pressão osmótica, ambos dependentes do teor de nanopartículas no
nanocompósito.
Por fim, realizaram-se estudos de libertação do azul de metileno em
nanocompósitos seleccionados, à temperatura fisiológica. Os perfis de
libertação obtidos foram analisados segundo os modelos de Korsmeyer-
Peppas, Higuchi, Hixson-Crowell e cinéticas de ordem zero e de primeira
ordem. De um modo geral o modelo de Korsmeyer-Peppas ajustou-se bem
aos resultados experimentais e permitiu inferir sobre o mecanismo de
libertação. In this work thermosensitive hydrogel nanocomposites of gelatin and k- carrageenan containing gold nanoparticles as the disperse phase were prepared and characterized. The study of the release kinetics of a model drug was carried out. The effect content and morphology (nanospheres or nanorods) of the gold nanoparticles on the optical, thermal, swelling and mechanical properties of the nanocomposites was studied. The nanocomposites showed the optical properties of the nanoparticles used as dispersed phase. It was observed that the addition of nanoparticles to the k- carrageenan nanocomposites increases the melting temperature and the elastic modulus of the gel, which means that the nanoparticles act as a reinforcement of the polymer matrix. However, in the case of the gelatin nanocomposites, an opposite behavior was observed suggesting that the nanoparticles inhibit the gelatin crosslinking of the gelatin. The swelling tests suggest that the nanocomposites swelling depend on the gel “strength” and on the osmotic pressure effect, both effects depending on the nanoparticles content of the nanocomposite. Finally release studies of methylene blue as a model drug were carried out at the physiological temperature. The release profiles were analyzed using the Korsenmeyer-Peppas model, Higuchi model, Hixson-Crowell model and zero order and first order kinetics equations. Overall the Korsmeyer-Peppas model adjusted well to the experimental data and allowed to conclude about the release mechanism. |
