Efeitos terapêuticos de Inula crithmoides em ratinhos expostos ao CCI4

Abstract

A actividade regeneradora e protectora do extracto aquoso de porções aéreas de Inula crithmoides L. foi avaliada em ratinhos expostos ao CCl4. O fígado, rim e baço foram analisados histologicamente para verificar estas actividades. Os dados histológicos mostraram que o extracto aquoso não apresentou toxicidade na dose de 250 mg/Kg e que possuía um efeito hepatoregenerador nas doses administradas (250 mg/Kg e 500 mg/Kg). Outro objectivo do presente estudo, consistiu em avaliar a actividade regeneradora das fracções de acetona, etanol, etanol:água (50:50) e resíduo do extracto aquoso de I. crithmoides. Posteriormente, foram isolados por cromatografia em coluna e caracterizados por análise espectroscópica, os constituintes químicos da fracção que apresentou um maior efeito regenerador. Os dados histológicos revelaram que a fracção de etanol:água (50:50) (250 mg/Kg) foi a que apresentou uma maior actividade regeneradora, principalmente no fígado. Os compostos isolados foram o ácido 3,4-diacetoxi-cinâmico (1) e o dissacarídeo 4-O-[6’-(1’,2’,3’,4’-tetra-O-acetil)frutofuranosil]-1,2,3,6-tetra-O- acetilfrutofuranose-D (2). Os resultados obtidos sugerem que o extracto aquoso não revelou um efeito protector. Apresentou um efeito hepatoregenerador e a fracção de etanol:água (50:50) foi a que revelou uma maior actividade regeneradora. A realização do presente estudo, leva-nos a concluir que é importante aprofundar o conhecimento das propriedades terapêuticas de I. crithmoides, com vista à sua futura aplicação, e realizar estudos de isolamento e caracterização dos seus princípios activos.

The regenerative and protective activity of aqueous extract of aerial parts of Inula crithmoides L. was evaluated in mice exposed to CCl4. The liver, kidney and spleen were histologically analyzed to verify these activities. Histological analysis revealed that the aqueous extract was not toxic at a dose of 250 mg/kg and revealed hepatoregenerative effects in the dosage administered (250 mg/kg and 500 mg/Kg). Another aim of this study, was to evaluate the regenerative activity of the fractions of acetone, ethanol, ethanol:water (50:50) and residue of aqueous extract of I. Crithmoides. Were isolated by column chromatography and characterized by spectroscopic analysis the chemical constituents of the fraction showed a greater regenerative effect. The histological data revealed that the fraction of ethanol:water (50:50) (250 mg/Kg) showed the greater regenerative activity, mainly in the liver. The compounds isolated were 3,4-diacetoxycinnamic acid (1) and 4-O-[6’-(1’,2’,3’,4’-tetra-O-acetyl)fructofuranosyl]-1,2,3,6-tetra-O-acetylfructofuranose - D (2). The results obtained suggest that the aqueous extract didn’t show a protective effect. However, a hepatoregenerative effect is suggested, and the fraction of ethanol: water (50:50) showed greater regenerative activity. This study, leads us to conclude that it is important to better understand the knowledge of the therapeutic properties of I. crithmoides for future use and, perform studies of isolation and characterization of its active principles.
