Abstract: | O principal objectivo da presente dissertação de mestrado é a optimização da
produção de compósitos de nitreto de silício (Si3N4)-biovidro e avaliação do
respectivo desempenho mecânico e biológico tendo em vista a sua utilização
em aplicações biomédicas.
Este biomaterial é composto por 70% e 30% em peso de Si3N4 e biovidro,
respectivamente, combinando vantajosamente as propriedades de ambos os
constituintes. O Si3N4 é um material que apresenta uma excelente conjugação
de propriedades mecânicas para um cerâmico e funciona como matriz
estrutural deste biocompósito. O biovidro confere-lhe bioactividade,
promovendo igualmente a sinterização na presença de fase líquida. A
produção de amostras com 98% de densidade relativa foi possível através da
optimização das condições de prensagem a quente. Após alguns estudos
neste âmbito, utilizou-se o seguinte ciclo de sinterização: velocidade de
aquecimento de 10ºC/min até 1350ºC, seguido por um patamar de 40 min, sob
uma pressão de 30 MPa. A utilização deste ciclo permitiu velocidades de
densificação elevadas evitando a cristalização da fase intergranular,
constituída pelo biovidro, mantendo desta forma as suas propriedades
intrínsecas. A caracterização microestrutural do compósito e os estudos da
cinética de sinterização permitiram concluir que a densificação ocorreu
essencialmente durante o primeiro estágio da prensagem a quente assistida
pela fase líquida. A existência de uma quantidade significativa desta fase
possibilitou o rearranjo das partículas de Si3N4 e apenas uma pequena parte
da densificação é acompanhada pela transformação de fase α-Si3N4→β-Si3N4.
O compósito Si3N4-biovidro apresenta tipicamente as seguintes propriedades
mecânicas: tenacidade à fractura, KIC = 4.4±0.5MPa.m1/2; dureza Vickers, HV =
10.3±0.5GPa; módulo de Young, E = 197±4GPa; resistência à flexão, σR =
383±47MPa; e finalmente, o módulo de Weibull, m = 8.3±0.6. A realização de estudos de molhabilidade com vários líquidos, água,
diiodometano, SBF (simulated body fluid) e uma solução de albumina bovina
(SBF+BSA-bovine serum albumin), permitiu caracterizar a superfície deste
novo compósito. A 25ºC, revelou um carácter particularmente hidrofílico sendo
o valor do ângulo de contacto com a água, 26.6±2.0º, e o valor de tensão
superficial, γSV = 66.6 mJ.m-2, superior ao dos biocerâmicos mais comuns.
Outra característica importante é o elevado trabalho de adesão, Wa=96.4±0.2
mJ.m-2, que apresenta em relação à solução proteica SBF+BSA. Estudos de
potencial zeta permitiram concluir que para valores de pH superiores ao ponto
isoeléctrico, pHIEP=2.5, o compósito Si3N4-biovidro encontra-se carregado
negativamente. Isto deve-se, principalmente, à presença de ligações Si-O
insaturadas e grupos Si-OH na sua superfície. Desta forma, quando imerso na
solução SBF+BSA, que apresenta um valor de pH igual a 7, o compósito é
fortemente negativo. A adsorção da albumina nestas condições explica-se em
termos de ganhos de entropia e entalpia resultantes da sua desnaturação e
também através da coadsorção de catiões na superfície do material.
O tratamento do compósito Si3N4-biovidro, através da sua imersão em meio de
cultura celular, demonstrou que a adsorção de proteínas é um processo que
ocorre rapidamente na superfície do material. Verificou-se igualmente a
dissolução da fase intergranular ao longo do tempo, acompanhada da
formação de cavidades de várias dimensões. A avaliação da proliferação
celular durante um período de cultura de 5 dias permitiu concluir que as
superfícies, com e sem pré-tratamento, apresentam um comportamento
semelhante em relação à proliferação da linha celular de osteoblastos
humanos MG63. The main objective of this master thesis is to optimize the production of silicon nitride (Si3N4)-bioglass composites and to determine the mechanical and biological behavior in order to assess their potential for biomedical applications. This biomaterial is composed by 70wt.% Si3N4 ceramic phase and 30wt.% of bioglass, advantageously combining the properties of each constituent. The Si3N4 presents outstanding mechanical properties becoming the structural matrix of the biocomposite. The bioglass provides bioactivity and simultaneously assists on the liquid phase sintering of the Si3N4 grains. The production of samples with 98% of relative density was possible through the optimization of the hot pressing conditions. After some studies the following sintering cycle was selected: heating rate of 10ºC/min up to 1350ºC, followed by a stage of 40 min at the dwell temperature under a pressure of 30MPa. The use of this cycle allowed a very fast densification rate, avoiding the crystallization of the bioglass intergranular phase and therefore maintaining its intrinsic properties. The microstructural characterization of the composite together with the sintering kinetics evaluation showed that the densification essentially took place during the first stage of the liquid phase sintering. The large amount of glassy phase assures the densification by liquid phase assisted Si3N4 grains rearrangement and only a residual parcel of the densification is due to the phase transformation α-Si3N4→β-Si3N4. The Si3N4- bioglass composite presents the following mechanical properties: fracture toughness, KIC = 4.4±0.5MPa.m1/2; Vickers hardness, HV = 10.3±0.5GPa; Young’s modulus, E = 197±4GPa; bending strength, σR = 383±47MPa; and finally, Weibull modulus, m = 8.3±0.6. Wettability studies performed with several liquids, water, diiodomethane, SBF (simulated body fluid) and a bovine albumin solution (SBF+BSA-bovine serum albumin), allowed the characterization of the composite surface properties. At 25ºC, the Si3N4-bioglass composite reveals a hydrophilic character with a water contact angle value of 26.6±2.0º, and a surface tension value, γSV = 66.6 mJ.m- 2, comparatively higher than those of the most common bioceramics. Another important feature is the high work of adhesion towards SBF+BSA, Wa=96.4±0.2 mJ.m-2. Zeta potential studies showed that the Si3N4-bioglass composite is negatively surface charged above the isoelectric point, pHIEP=2.5. This is due to the presence at the surface of unsaturated Si-O bonds and Si- OH functional groups. Therefore, the composite surface is strongly negative when immersed in the solution SBF+BSA, which presents a pH value of 7. Albumin adsorption, in these conditions, is explained in terms of entropy and enthalpy gains from conformational unfolding and cations coadsorption. The treatment of the Si3N4-bioglass composite with culture medium revealed that the protein adsorption is an early process that happens in the material surface. It was also verified that some dissolution of the intergranular phase occurs, which showed to be time-dependent, with the formation of different size cavities. The evaluation of the cell proliferation, during a 5 days culture period, shows that the “as-received” and pretreated surfaces present a similar behaviour concerning the proliferation of MG63 osteoblast-like cells. However, for the same culture period, the cell cultures carried out on the Si3N4-bioglass composite surface present a superior cell growth comparing to the control cultures (standard plastic culture plates), revealing a possible inductive effect in the proliferation of the MG63 cells. No evidence of cytotoxic responses was observed. |
