Abstract: | Myotonic Dystrophy type 1 (DM1) is the most common muscular dystrophy
type among adults being an incurable disease with no approved treatment.
The aim of the present study is to identify novel molecular targets related to
protein phosphorylation alterations observed in patients with DM1, using an
unbiased strategy of automatic text mining traditionally applied in
bibliometric studies. Using the Scopus database, 1303 manuscripts
combining DM1 and protein phosphorylation, were identified. However,
only 713 met the desired criteria. These were analysed using the VOSviewer
software, through which networks of keywords associated with DM1 were
created, allowing visualising the terms that have been well correlated with
the disease and those that are still lacking scientific evidence. Interestingly,
protein phosphorylation was one of the evidenced molecular mechanisms
associated to DM1. Through this approach a list of genes associated with
DM1 was collected, which comprises several proteins associated with protein
phosphorylation. Through the Venn diagram, the genes identified in the
bibliometric analysis were compared with the genes already associated with
DM and DM1, present in the DisGeNET platform. The majority of the genes
identified in present study proved to be novel in the context of DM1
pathology were not previously associated to DM or DM1 in DisGeNET. In
addition, a functional enrichment analysis of the groups of genes compared,
revealed significant differences in both their characteristics and biological
functions. This reflects the lack of studies on protein phosphorylation in
DM1, suggesting that this process can contribute to a better characterization
of the disease. In this sense, some molecular targets associated to protein
phosphorylation were chosen for validation and the intracellular levels of
these proteins, were evaluated, using DM1patient-derived fibroblasts. The
APP, ERK1 and AKT1 showed significant increases in DM1-patient derived
fibroblasts when compared with controls, as well as PP1γ, PP2B. Given the
evident dysregulation in the phosphoproteins, kinases and phosphatases
evaluated here, the hypothesis that protein phosphorylation may be a crucial
process in the development of DM1 was strengthened and further studies are
welcome to further explore these phosphotargets and propose a therapy for
DM1. A Distrofia Mitónica do tipo 1 (DM1) é o tipo de distrofia muscular mais comum em adultos, sendo uma doença incurável, sem tratamento aprovado. O objetivo do presente estudo é identificar novos alvos moleculares relacionados com as alterações na fosforilação de proteínas observadas em doentes com DM1, utilizando uma estratégia imparcial de mineração automática de texto tradicionalmente aplicada a estudos bibliométricos. Utilizando a base de dados Scopus, identificaram-se 1303 manuscritos que combinam a DM1 e a fosforilação de proteínas. Contudo, apenas 713 cumprem os critérios desejados. Estes, foram analisados com recurso ao software VOSviewer, através do qual se criaram redes de palavras-chave associadas à DM1, permitindo visualizar os termos que mais têm sido correlacionados com a doença e aqueles que carecem de evidências científicas. Curiosamente, a fosforilação de proteínas foi um dos mecanismos moleculares evidenciados como associados a DM1. Através desta abordagem recolheu-se uma lista de genes associados à DM1, que compreende várias proteínas associadas à fosforilação de proteínas. Através do diagrama de Venn, compararam-se os genes identificados na análise bibliométrica, com os genes já associados à DM e DM1, presentes na plataforma DisGeNET. A vasta maioria dos genes identificados no presente estudo revelaram ser novidade, no contexto da patologia, não foram previamente associados à DM ou DM1 no DisGeNET. Complementando, realizou-se uma análise de enriquecimento funcional, dos grupos de genes comparados, que revelou haver diferenças significativas tanto nas características como nas funções biológicas dos mesmos. Isto traduz a falta de estudos sobre a fosforilação de proteínas em DM1, sugerindo que este processo pode contribuir para uma melhor caracterização da doença. Neste sentido, alguns alvos moleculares associados à fosforilação de proteínas foram escolhidos para validação e os níveis intracelulares dessas proteínas, foram avaliados, usando amostras de fibroblastos provenientes de doentes com DM1. A APP, ERK1 e AKT1 mostraram aumentos significativos em fibroblastos derivados de pacientes com DM1 quando comparados com os controlos, assim como PP1γ, PP2B. Perante a evidente desregulação nas fosfoproteínas, nas quinases e fosfatases aqui avaliadas, a hipótese que a fosforilação de proteínas pode ser um processo crucial no desenvolvimento do DM1 foi reforçada, pelo que são bem-vindos mais estudos exploratórios destes alvos de fosforilação e propor uma terapia para DM1. |
