Abstract: | A oxitetraciclina é um dos antibióticos mais usados no tratamento de
peixes de aquicultura na Europa. A presença de elevadas quantidades
deste antibiótico em águas de aquicultura pode levar à contaminação dos
recursos hídricos e ao aparecimento de resistência bacteriana. O
objetivo deste trabalho foi o desenvolvimento de um sensor de massa
para detetar a oxitetraciclina (OTC) nas águas da aquicultura, tendo por
base os cristais de quartzo piezoelétricos (QCM) com polímeros
molecularmente impressos (MIP) como elemento de reconhecimento.
Sintetizaram-se quatro polímeros molecularmente impressos (MIP-1,
MIP-2, MIP-3, MIP-4) descritos na literatura para a deteção da OTC. Para
imobilizar os MIPs na superfície do cristal de quartzo, estes foram
introduzidos numa membrana de PVC e plastificante, dissolvida em
tetrahidrofurano (THF), que posteriormente foi colocada na superfície de
ouro do elétrodo do cristal piezoelétrico. Os cristais depois de revestidos
e secos foram avaliados num sistema de injeção em fluxo (FIA). A injeção
de soluções padrão com diferentes concentrações da oxitetraciclina e
dos seus possíveis interferentes, permitiu construir as várias curvas de
calibração e estudar o comportamento do sensor. O objetivo era
perceber qual o polímero molecularmente impresso que melhor se
adequa para a deteção da OTC.
Os sensores revestidos com o MIP-1 e o MIP-2 foram os que se
mostraram ser mais seletivos para a OTC face à tetraciclina (TC), à
ciprofloxacina (CIP), à enrofloxacina (ENR) e ao ácido oxolínico (AO). No
entanto o MIP-2 respondeu mais ao NaCl do que o MIP-1, o que torna a
sua aplicação desaconselhada em águas estuarinas
O sensor revestido com o MIP-1 apresenta uma sensibilidade para a
OTC em tampão HEPES de 502275 Hz L mol-1, um limite de
quantificação (LOQ) de 4,91 ppm e um limite de deteção (LOD) de 1,47
ppm. Contudo estes valores podem ser otimizados no futuro, otimizando
o revestimento do sensor, na quantidade, disposição de MIP e o modo
como este se encontra incorporado do PCV. Oxytetracycline is one of the most widely used antibiotics in the treatment of aquaculture fish in Europe. The presence of high amounts of this antibiotic in aquaculture waters can lead to contamination of water resources and the appearance of bacterial resistance. The main goal of this work was the development of a mass sensor to detect oxytetracycline (OTC) in aquaculture waters, based on piezoelectric quartz crystals (QCM) with a molecularly imprinted polymers (MIP) as a recognition element. Four molecularly imprinted polymer (MIP) recipes existing in the literature, for OTC measurement were synthesized (MIP-1, MIP-2, MIP 3, MIP-4). These four MIP were immobilized on the surface of the quartz crystal, by introduced them into a plasticized PVC membrane dissolved in tetrahydrofuran (THF), which was later placed on the gold surface of the piezoelectric crystal electrode. The crystals, after being coated and dried, were evaluated in a flow injection system (FIA). The injection of standard solutions with different concentrations of oxytetracycline or possible interfering compounds, allowed to build the various calibration curves and study the behaviour of the sensor. The goal was to understand which molecularly imprinted polymer best suits for OTC detection. The sensors coated with MIP-1 and MIP-2, were the ones that proved to be more selective for OTC compared to the possible interfering compounds, tetracycline (TC), ciprofloxacin (CIP), enrofloxacin (ENR) and oxolinic acid (AO). However, MIP-2 responded more to NaCl than MIP-1, which discourages its application in estuarine waters. The sensor coating with MIP-1, has an OTC sensitivity in HEPES buffer of 502275 Hz L mol-1, a quantification limit (LOQ) of 4,91 ppm and a detection limit (LOD) of 1,47 ppm. These values can be optimized in the future, optimizing the sensor coating, in quantity, disposition on the quartz crystal and the way it is incorporated in the PVC. |
