ABSTRACT: The search of new biologically active compounds and consequently possible use in the treatment of several diseases has been improving over the last decades. Among the target compounds submitted to an intensive research, benzodiazepines play an important role. This dissertation has as objective the synthesis of new 1,5-benzodiazepines, which have been reported as an important class of benzodiazepines. In the first part of this work, a review on the chemical structure and numbering system, natural occurrence, some methodologies of synthesis and their applications are presented. The second part describes the synthesis of 4-aryl-2-(2-hydroxyphenylethylidenone)- 3,4-dihydro-1H,5H-benzo[b][1,5]-diazepines, 2-(2-hydroxyphenyl)-4-styryl-3Hbenzo[ b][1,5]-diazepines, and 1-acetyl-2-(2-hydroxyphenyl)-4-styryl-1H-benzo [b][1,5]-diazepines, obtained by the reaction of 5-aryl-3-hydroxy- 1-(2-hydroxyphenyl)-2,4-pentadiene-1-one with ortho-phenylenediamine. The former have been obtained from the conjugated addition of orthophenylenediamine to the α,β-insaturated carbonyl moiety while the latter were obtained by the condensation of ortho-phenylenediamine with β-diketone moiety. The 5-aryl-3-hydroxy-1-(2-hydroxyphenyl)-2,4-pentadiene-1-ones were previously prepared from the Baker Venkataraman rearrangement of 2-cinnamoyloxyacetophenones. Besides the desired benzodiazepines, some by products have been obtained, being the 2-styrylchromones the most important ones. In the third part the structural characterization of the new 1,5-benzodiazepines and other new compounds are described in detail. For that, we used exhaustive nuclear magnetic resonance (NMR) spectroscopy studies, including 1H, 13C, 2D-NMR homo- and heteronuclear correlated spectroscopy (HSQC and HMBC).
