Title: The use of imposex for monitoring TBT pollution in the Portuguese coast, with emphasis on the gastropod Nassarius (Hinia) reticulatus (L.)

Abstract: TBT pollution started in the mid-1960s with the use of tributyltin (TBT) antifouling paints on ships. Its deleterious effects on non-target organisms became apparent in the 1970s, with the upsurge of oyster shell thickening and of prosobranch (snails) females with male characters (imposex) throughout the world. In the early 1980s it was recognised that TBT pollution was the cause of those effects and legislation that restrics the use of TBT paints was soon introduced in several countries. After the mid 1980s many studies have described the TBT toxicity on organisms over a broad taxonomic spectrum, from bacteria to vertebrates, and its negative impacts at the individual, populational and community levels. Methods were developed for biomonitoring TBT pollution, based on specific biomarkers (mainly the imposex), and biomonitoring surveys were carried out along extensive coastal areas around the world. In the 1990s it was recognised that the existing legislation was not effective to reduce TBT pollution at a global scale and it was proposed that a total ban on TBT and other organotin compounds from antifouling paints should take place during the first decade of the 2000 millennium. The present work resulted from the need to monitor TBT pollution on the Portuguese coast, considering the very scarce information that was available for this particular area. The studies performed under the scope of the current dissertation enabled to define guidelines for monitoring the Portuguese inshore coast. It is proposed that biomonitoring along the seashore should be based on the prosobranch species Nassarius reticulatus (L.) and Nucella lapillus (L.). For monitoring the innermost portions of the estuarine systems, the low salinity tolerant species Hydrobia ulvae (Pennant) should also be used. The first species, N. reticulatus, is the most important bioindicator in view of its abundance over the entire mainland coast and its adequate properties for describing TBT gradients around hotspots. As a matter of fact, at the Ria de Aveiro, N. reticulatus provided the best description of the TBT gradients that occur between the sea and the ria and also inside the estuarine system in comparison to other bioindicators, like N. lapillus, H. ulvae and Littorina littorea (L.). For this reason, important aspects of its biology were studied in order to define the methodology to be applied for monitoring the national mainland coast.The reproductive cycle of Nassarius reticulatus was investigated at the Ria de Aveiro and a clear seasonal pattern was observed, with the beginning of gametogenesis in August followed by ripening of the gonads in January and February and spawning from February to July. A considerable variation of the male penis length was observed along this cycle, with maximum values during the ripening of the gonads in the winter and minimum values in the rest phase during the summer. Consequently, the imposex index “RPLI” (relative penis length) also varies seasonally and thus it can only be used for imposex comparisons between populations at approximately the same phase of the reproductive cycle. As the cycle appears to be synchronised along the Portuguese coast, it is recommended that the biomonitoring surveys should be carried out during a short period (2-3 months) in the same time of the year. Some aspects related to the individual growth of Nassarius reticulatus were also studied at the Ria de Aveiro. It was found that growth occurs during the first five years of life and that the longevity probably reaches 11 or more years. The growth rate is nearly null around the winter and is higher in the summer. Nevertheless, the shell rings are rarely formed and thus they can not be used to estimate the age of the snails. Sexual maturation is attained between the 3rd and 4th years in males and between the 4th and 5th years in females, while imposex is initiated at least within the 2-3 years of age. Considering the high longevity of the species, the irreversibility of the imposex development and the difficulty to age the animals by shell rings counting, it is recommended that biomonitoring surveys should be repeated at time intervals of about 5 years in order to detect a putative decline of the pollution levels in the future. The effectiveness of some compounds derived from antifouling paints to promote imposex in Nassarius reticulatus was tested under laboratory conditions. It was shown that, besides TBT, triphenyltin (TPT) can also induce imposex in this species. This is a very relevant aspect to be taken into account for a correct interpretation of the imposex levels registered in the monitoring surveys. For further validation of Nassarius reticulatus as a TBT indicator species the imposex/TBT body burden relationships determined for the Portuguese coast were compared with others reported in the literature for other sites in Europe. A considerable variation was found in this relationship along the European coast. Consequently, it is recommended that monitoring programmes should include both imposex measurements and chemical analysis of organotin contamination in the female tissues and, whenever possible, in the environment. Biomonitoring surveys performed in 2000 have shown that TBT pollution in the Portuguese coast is higher at estuarine systems than at the open coast. This is due to the fact that ports, dockyards and most marinas are located inside the estuarine areas. At these sites Nassarius reticulatus populations exhibited TBT female body burdens in the range of 100-1400 ng Sn g-1 dw and very high values of imposex, resembling those reported by other authors for the most polluted harbours of Spain, France and England: RPLI=20-90%; VDSI (vas deferens sequence index)=2-5. Sterile females were found at some Portuguese harbours, a phenomenon possibly associated to TBT contamination since it occurred only in females heavily affected by imposex. Nucella lapillus was only found at sites of the open coast and, for this reason, the surveyed populations presented low to moderate levels of TBT contamination in the female tissues and of imposex: <20-147 ng TBT-Sn g-1 dw; RPSI (relative penis size)=0-33%; VDSI=1.4-4.1; %S (percentage of sterile females)=0-8%. The absence of N. lapilus from harbours is probably linked to local extinction after female sterilisation by TBT. For instance, at the Ria de Aveiro this species exhibited increasing levels of imposex and of female sterility from the sea into the ria and this trend was accompanied by a drastic decrease of abundance. Time comparisons of the available data on N. lapillus imposex in the Portuguese coast suggest a global increase of TBT pollution in recent years. This fact indicates that the existing legislation failed in the aim of reducing the pollution levels and, therefore, supports the need to eliminate the use of TBT (and TPT) in antifouling paints, as planned in the “International Convention on the Control of Harmful Antifouling Systems on Ships” approved in October 2001 by IMO (International Maritime Organisation). The implementation of regular monitoring programmes gains further relevance if we want to evaluate the effectiveness of the legislative measure referred above. Since the OSPAR (Convention for the Protection of the Marine Environment of the North-East Atlantic) guidelines for TBT monitoring do not adequately suit the conditions prevailing in the Portuguese coast, mainly in what regards the selection of the indicator species, it is proposed that they should be improved in some key aspects in accordance to the conclusions derived from the present work.
A poluição marinha por tributilestanho (TBT) surgiu em consequência da utilização de tintas antivegetativas com TBT para pintar os cascos de embarcações, iniciada em meados da década de 1960. Os efeitos deste tipo de poluição manifestaram se na década de 70, em vários locais do mundo, com o aparecimento de ostras com conchas muito espessas e de fêmeas de gastrópodes prosobrânquios (búzios) com características masculinas (imposex). No início da década de 80 identificou-se a contaminação ambiental por TBT como causa destes fenómenos e adoptaram-se as primeiras medidas legislativas para restringir a utilização de tintas contendo este tipo de compostos. A partir de meados dos anos 80 comprovou-se a toxicidade do TBT para uma grande diversidade de organismos, desde bactérias a vertebrados, bem como os seus efeitos negativos aos níveis individual, populacional e da comunidade. Desenvolveram-se métodos para a biomonitorização deste tipo de poluição com base em biomarcadores específicos, principalmente o imposex, e foram avaliados os níveis de poluição da zona costeira em vastas áreas geográficas do mundo. Na década de 90 constatou-se que a legislação vigente tinha uma reduzida eficácia no combate a este tipo de poluição. Foi, então, proposta a abolição total da utilização de tintas com TBT, para ser implementada na primeira década de 2000. O presente trabalho surgiu da necessidade de se monitorizar a poluição por TBT na costa portuguesa, dada a escassa informação até há pouco tempo existente para esta área geográfica. A partir dos estudos desenvolvidos no âmbito desta tese, foi possível definir metodologias para a monitorização da zona costeira com base na aplicação combinada de três espécies de prosobrânquios, utilizando o imposex como principal biomarcador. Propõe-se que a biomonitorização ao longo da costa incida sobre Nassarius reticulatus (L.) e Nucella lapillus (L.), podendo adicionalmente ser utilizada a espécie Hydrobia ulvae (Pennant) nas regiões de menor salinidade no interior dos sistemas estuarinos. A espécie indicadora mais importante é N. reticulatus, devido à sua ampla distribuição geográfica na costa portuguesa associada a uma sensibilidade adequada para descrever gradientes de contaminação em torno de “pontos quentes” de poluição. De facto, na Ria de Aveiro, onde existem importantes fontes de poluição por TBT, N. reticulatus foi a espécie que melhor descreveu os gradientes de contaminação entre o mar e a ria e os existentes no interior do sistema estuarino, em comparação com outras espécies indicadoras, designadamente N. lapillus, H. ulvae e Littorina littorea (L.). Assim, foram estudados alguns aspectos da biologia de N. reticulatus com vista a uma melhor definição da metodologia a aplicar nas campanhas de biomonitorização da costa continental portuguesa. Estudou-se o ciclo reprodutor desta última espécie na Ria de Aveiro e registou-se um padrão de variação sazonal pronunciado neste ciclo, com o início da gametogénese em Agosto, a maturação das gónadas em Janeiro-Fevereiro e a desova entre Fevereiro e Julho. Verificou-se que o comprimento do pénis dos machos varia ao longo do ciclo, atingindo valores máximos na época de maturação dos testículos (Inverno) e mínimos na época de repouso sexual (Verão). Consequentemente, o índice de imposex “RPLI” (comprimento relativo do pénis) deverá apresentar também variações sazonais, pelo que a comparação dos valores de “RPLI” entre populações diferentes só é válida se estas se encontrarem no mesmo estádio do ciclo reprodutor. Visto que este ciclo parece estar sincronizado ao longo da costa portuguesa, recomenda-se que as campanhas de monitorização sejam efectuadas num curto período de tempo (2-3 meses) e sempre na mesma época do ano. Estudaram-se, também, alguns aspectos relacionados com o crescimento individual e a longevidade de Nassarius reticulatus na Ria de Aveiro. Verificou-se que o crescimento ocorre nos primeiros 5 anos de vida, embora a longevidade tenha sido estimada em, pelo menos, 11 anos. A taxa de crescimento apresenta uma forte variação sazonal, com valores mais elevados no Verão e nulos no Inverno. Apesar disso, nem sempre ocorre a correspondente formação de anéis de crescimento, pelo que a sua contagem não possibilita a determinação da idade. A maturação sexual é atingida aos 3-4 anos de vida, no caso dos machos, e aos 4-5 anos, no caso das fêmeas. O desenvolvimento do imposex inicia-se, pelo menos, aos 2-3 anos de idade. Tendo em conta a irreversibilidade do desenvolvimento do imposex, a elevada longevidade da espécie e a dificuldade em determinar a idade dos animais, recomenda-se que as campanhas de biomonitorização sejam realizadas com uma periodicidade de cerca de 5 anos para se poder detectar um eventual declínio dos níveis de poluição. Testou-se em laboratório a acção de vários compostos derivados das tintas antivegetativas na indução de imposex em Nassarius reticulatus. Verificou-se que o trifenilestanho (TPT), para além do TBT, também promove o desenvolvimento de imposex nesta espécie, sendo esta informação relevante para a correcta interpretação dos níveis de imposex observados nas populações deste gastrópode. Com vista a uma completa validação de Nassarius reticulatus como espécie indicadora, estudou-se também a relação entre os níveis de imposex e a contaminação dos tecidos das fêmeas por TBT, tendo-se verificado, por comparação com trabalhos de outros autores, que esta relação pode apresentar uma variação acentuada ao longo da costa europeia. Este facto realça a necessidade de que os estudos de imposex sejam acompanhados por medições analíticas dos compostos orgânicos de estanho nos tecidos das fêmeas e, se possível, também no ambiente. Campanhas de biomonitorização realizadas no ano de 2000 mostraram que os níveis de poluição por TBT na costa ocidental portuguesa apresentam valores mais elevados no interior de sistemas estuarinos e valores mais reduzidos na costa aberta. Esta situação resulta do facto dos principais portos, estaleiros navais e marinas se localizarem no interior de estuários. As populações de Nassarius reticulatus nestas áreas apresentaram níveis de contaminação por TBT nos tecidos da ordem dos 1001400 ng TBT-Sn g-1 (peso seco) e níveis muito elevados de imposex, comparáveis aos descritos por outros autores para os principais portos de Espanha, França e Inglaterra: RPLI=20-90%; VDSI (desenvolvimento do vaso deferente)=2-5. Em algumas áreas portuárias foram encontradas fêmeas esterilizadas, sendo este fenómeno uma possível consequência do desenvolvimento de imposex. Por sua vez, Nucella lapillus foi apenas encontrada em locais da costa aberta, razão por que apresentou níveis baixos ou moderados de contaminação por TBT e de imposex: <20-147 ng TBT-Sn g-1 (peso seco); RPSI (tamanho relativo do pénis)=0-33%; VDSI=1,4-4,1; %S (percentagem de fêmeas estéreis)=0-8%. O facto de não se ter encontrado N. lapillus no interior das zonas portuárias poderá estar relacionado com a sua extinção nesses locais, possivelmente devida à esterilização das fêmeas em consequência de uma elevada contaminação por TBT. Esta situação foi observada na Ria de Aveiro, onde um gradiente crescente de contaminação da zona costeira para o interior da ria foi acompanhado por um aumento da percentagem de fêmeas esterilizadas e por uma diminuição drástica da abundância da espécie. A comparação dos níveis de imposex de N. lapillus em 2000 com os descritos no passado recente por outros autores sugere que ocorreu um aumento da poluição por TBT na costa portuguesa. Este facto revela a reduzida eficácia da legislação vigente no combate a este tipo de poluição e suporta a necessidade de se eliminar por completo a utilização de tintas antivegetativas com TBT (e TPT) a uma escala global, conforme previsto na Convenção Internacional sobre o Controlo da Utilização de Sistemas Antivegetativos Nocivos em Embarcações, aprovado pela IMO (Organização Marítima Internacional) em Outubro de 2001. A monitorização da poluição por TBT assume uma importância crucial no futuro se se pretender avaliar a eficácia da medida legislativa acima referida. Tendo em consideração que as linhas orientadoras para a biomonitorização deste tipo de poluição definidas pela OSPAR (Convenção para a Protecção do Ambiente Marinho do Nordeste Atlântico) não se adequam à costa portuguesa, essencialmente no que diz respeito à selecção das espécies indicadoras, propõe-se que elas sejam melhoradas em alguns aspectos, de acordo com os resultados obtidos no presente trabalho.
