Abstract: | A telomerase é a enzima responsável pela proliferação
descontrolada de células tumorais. A estabilização, por
ligandos, das estruturas de G-quadruplexes de ADN tem
vindo a ser apontada como uma estratégia promissora na
inibição da telomerase em terapias de cancro. Neste trabalho
foram estudadas várias porfirinas e ftalocianinas catiónicas
como potenciais ligandos estabilizadores de G-quadruplexes
de ADN.
Selecionou-se, como padrão, uma das porfirinas mais
estudadas nesta área de investigação, a meso-tetraquis(Nmetilpiridínio-
4-il)porfirina (TMPyP), que apresenta como forte
limitação a sua baixa seletividade para G-quadruplexes. Os
estudos de afinidade e seletividade dos ligandos
selecionados recorrendo a técnicas de espectroscopia de
ultravioleta-visível mostraram que, quer a coordenação do
núcleo da porfirina TMPyP com o ião metálico zinco(II) quer a
presença de unidades de piridina cationizadas nas posições
beta pirrólicas contribuem fortemente para o aumento da
seletividade destes ligandos para G-quadruplexes.
Relativamente à serie de ftalocianinas selecionadas foi
possível concluir que a existência de um equilíbrio entre o
número e posição das cargas positivas na estrutura destes
ligandos é fundamental para a existência de seletividade para
estruturas de G-quadruplexes.
Os resultados dos estudos anteriores foram ainda
corroborados recorrendo a técnicas espectroscópicas de
fluorescência. Para os ligandos mais promissores foram
também feitos estudos preliminares sobre as alterações
conformacionais induzidas pela presença dos ligandos nas
estruturas dos G-quadruplexes, usando o dicroísmo circular.
Neste trabalho foi também desenvolvido um método de
screening competitivo que permite avaliar a afinidade e
seletividade para G-quadruplexes de novos ligandos de uma
forma rápida e económica. O desenvolvimento deste método
envolveu uma grande componente de otimização de
condições experimentais, sendo a sua utilização possível em
espetrofotómetros convencionais ou em espetrofotómetros
com leitor de placas. Telomerase is an enzyme responsible for the unlimited proliferation of cancer cells and the stabilization of DNA (deoxyribonucleic acid) G-Quadruplex structures has been pointed out as a promising strategy for telomerase inhibition in cancer therapies. In this work, several cationic porphyrins and phthalocyanines were studied as potential DNA GQuadruplexes stabilizing ligands. One of the most studied porphyrins in this field of research, the meso-tetrakis(N-methylpyridinium-4-yl)porphyrin (TMPyP) presents, as a strong limitation, low selectivity for GQuadruplexes. In these studies, TMPyP was selected as the standard. The studies of affinity and selectivity of selected ligands, using ultraviolet-visible spectroscopy techniques, have shown that either the coordination of the porphyrin TMPyP core with the zinc(II) metal ion or the presence of pyridine units cationized in the beta pyrrolic positions strongly contribute to increase the selectivity of these ligands to Gquadruplexes. With respect to the series of the selected phthalocyanines, it was possible to conclude that the existence of a balance between the number and position of the positive charges in the structure of these ligands is crucial for the existence of selectivity for G-quadruplex structures. Two of the studied phthalocyanines showed high selectivity and affinity to Gquadruplex structures, thus being identified as promising ligands in telomerase inhibition. The results of previous studies were further corroborated using spectroscopic fluorescence techniques. For the most promising ligands, preliminary studies of conformational changes induced by the presence of ligands in GQuadruplexes structures were also performed, using circular dichroism. In this work, a competitive screenning method that allows the evaluation of the affinity and selectivity of new ligands for Gquadruplexes, in a fast and economic way, was developed. The development of this methodology involved a large component of experimental conditions optimization, its application being proposed for conventional spectrophotometers or plate reader spectrophotometers. |
