Abstract: As macroalgas são uma fonte rica e equilibrada de nutrientes e fitoquímicos bioativos associados à prevenção de diversas doenças. Embora o consumo direto de macroalgas na Europa seja ainda muito insípido quando comparado com os países asiáticos, as indústrias alimentares mostram um crescente interesse em introduzir as macroalgas como ingrediente funcional em diversos produtos, antevendo-se um aumento significativo da inclusão de macroalgas nos hábitos alimentares nos países europeus. Neste trabalho pretendeu-se avaliar a potencialidade de extratos das macroalgas F. vesiculosus e Gracilaria sp. para utilização como ingrediente de alimentos e/ou suplementos. Para tal, foram preparados extratos aquosos (temperatura ambiente ou a 90ºC), acetona a 70% e etanol a 80%, que foram avaliados quanto ao seu teor de compostos fenólicos totais, atividade antioxidante (radical livre 2,2 difenil-1-picrilhidrazilo (DPPH•), poder antioxidante redutor férrico (FRAP) e co-oxidação do β-caroteno/ácido linoleico (β- caroteno)), capacidade de inibição de enzimas chave do metabolismo de carbohidratos (α-glucosidase, α-amilase) e de lípidos (lipase), bem como quanto ao seu valor nutricional no que respeita ao conteúdo em proteína, cinzas, fibra e açúcares. O extrato de etanol a 80% da F. vesiculosus apresentou o maior conteúdo de compostos fenólicos totais (5,7±0,3 g de equivalentes de ácido gálico/100g de extrato), e maior potencialidade nos ensaios de DPPH• (IC50= 99±7,2 μg/mL), FRAP (EC50= 0,29±0,03 mg/mL) e β-caroteno (IC50= 19,71±6,80 μg/mL). Por outro lado, no geral os extratos não mostraram grande potencial inibitório das enzimas em foco, com exceção do extrato de acetona a 70% da F. vesiculosus que apresentou um valor de IC50 de 0,032 mg/mL para a α-glucosidase. Em termos de valor nutricional, o extrato de água à temperatura ambiente da Gracilaria sp. revelou teores elevados de proteína (19±0,1%), cinza (55,9±1,0%) e fibra solúvel (14,9±0,8%). Por outro lado, os extratos de água quente e água à temperatura ambiente da F. vesiculosus apresentaram teores elevados de açúcares totais (13,8±2,1%) e fibra solúvel (17,2±0,7%), respetivamente. Os extratos aquosos da F. vesiculosus foram utilizados como ingredientes na formulação de alheiras vegetarianas numa percentagem de 0,4% (m/m peso húmido). As alheiras foram analisadas quanto ao teor de compostos fenólicos, atividade antioxidante e estado de oxidação pelos métodos Folin-Ciocalteu, Ácido 2,2´-azino-bis (3-etilbenzotiazolina-6-sulfónico) (ABTS•+) e substâncias reativas do ácido tiobarbitúrico (TBARS), respetivamente, bem como quanto aos seus conteúdos proteicos, cinzas, ácido lático e pH, parâmetro de cor e análise sensorial. Os resultados obtidos indicaram não existirem diferenças significativas entre as alheiras contendo o extrato de macroalga e a formulação base da alheira (controlo), à exceção da análise de cor em que a incorporação do extrato fez diminuir o parâmetro L* quando comparado com a alheira controlo.
Macroalgae are a rich and balanced source of bioactive nutrients and phytochemicals, associated with the prevention of various diseases. Although their direct diet consumption in Europe is still very low when compared to Asian countries, food industries have shown a growing interest in introducing macroalgae as a functional ingredient in diverse products, thus contributing to the increment of macroalgae in food habits in the European countries. The aim of this work was to evaluate the potential of macroalgae F. vesiculosus and Gracilaria sp. extracts as a food ingredient and/or supplements. F. vesiculosus and Gracilaria sp. extracts were produced with water (at room temperature or at 90°C), 70% acetone and 80% ethanol. The extracts were evaluated for their total phenolic compounds content, antioxidant activity ((2,2- diphenyl-1-picrylhydrazyl DPPH•), ferric reducing antioxidant power (FRAP) and co-oxidation of β-carotene/linoleic acid (β-carotene)), inhibition capacity of key enzymes of carbohydrate metabolism (α-glucosidase, α-amylase) and lipids (lipase), as well as their nutritional value in relation to the content of protein, ash, fiber and sugars. The 80% ethanol extract of F. vesiculosus was the most promising one with respect to the content of total phenolic compounds (5.7 ± 0.3 g of gallic acid equivalents / 100 g of extract), and antioxidant activity in DPPH• (IC50 = 99 ± 7.2 μg / mL), FRAP (EC50 = 0.29 ± 0.03 mg / mL) and β-carotene (IC50 = 19.71 ± 6.80 μg / mL) assays. Furthermore, the extracts did not show significant inhibitory potential of the enzymes in focus, with exception of the F. vesiculosus 70% acetone extract which showed an IC50 of 0.032 μg/mL towards the activity of α-glucosidase. Concerning the nutritional value, the room temperature water extract of Gracilaria sp. showed high levels of protein (19 ± 0.1%), ash (55.9 ± 1.0%) and soluble fiber (14.9 ± 0.8%). On the other hand, the hot water and room temperature extracts from F. vesiculosus exhibited high contents of total sugars (13.8 ± 2.1%) and of soluble fibers (17.2 ± 0.7%), respectively. The F. vesiculosus aqueous extracts were selected to be incorporated into vegetarian alheira at 0.4% (m/m wet weight), and were subsequently submitted to an evaluation of phenolic compounds, antioxidant activity and oxidation state by Folin-Ciocalteu, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS•+) and thiobarbituric acid reactive substances (TBARS) methods. Also protein, ash, lactic acid, pH, color and sensory analysis were evaluated. Results show that there were no significant differences between the alheiras containing the macroalgae extract and the base formulation of the alheira (control), except for the color analysis, in which the incorporation of the extract decrease the L * parameter against the control
