dc.description.abstractO osteossarcoma é o tumor maligno mais comum do osso, afetando particularmente adolescentes e jovens adultos, sendo o sexo masculino o mais afetado. O tratamento da doença nem sempre é eficaz, mas com a ajuda da quimioterapia aliada à remoção da parte afetada, a qualidade de vida dos doentes tem vindo a melhorar, verificando-se um aumento de sobrevivência. Este projeto surge com o intuito de desenvolver uma terapia combinada que venha a diminuir a resistência a quimioterápicos, utilizando compostos frequentemente usados na quimioterapia (etoposido e doxorrubicina) e um fitocomposto (hesperetina). Foi possível investigar os efeitos citotóxicos de fitocomposto isolado e em combinação na linha celular U2OS relativamente a diversos parâmetros celulares, nomeadamente a formação de colónias, proliferação celular, bem como a genotóxicidade e citótoxicidade. A formação de colónias foi seguida através do ensaio clonogénico, a proliferação celular foi determinada através dos ensaios de violeta de cristal (CV) e brometo de 3-[4,5-dimetil-tiazol-2-il]-2,5-difeniltetrazólio (MTT). O potencial genotóxico e citostático foi avaliado por ensaio de micronúcleos com bloqueio de citocinese (CBMN) e por análise citométrica do ciclo celular. Não ocorreram efeitos genotóxicos significativos, contudo a hesperetina isolada e em combinação induziu efeitos citostáticos, tal como observado por diminuição de índice de divisão nuclear (NDI) e alteração do perfil de fases do ciclo celular. Por comparação com agentes antineoplásicos, observou-se que também as combinações com hesperetina testadas não permitiram o desenvolvimento de colónias de U2OS. Estudos futuros de dano no ADN poderiam ilucidar na melhor compreensão das alterações que estes compostos provocam nesta linha celular e a comparação com outras linhas celulares também ajudaria na compreensão.pt
dc.description.abstractOsteosarcoma is the most common malignant tumor of the bone, occurring with more frequency in adolescents and young adults, with a greater percentage being male. The treatment is not always effective, but with chemotherapy as well as the removal of the affected section the quality of life of the patients has been increasing, together with the survival rate. This project has the aim of developing a combined therapy that can decrease the resistance to chemotherapeutics, utilizing compounds frequently used in chemotherapy (etoposide and doxorubicin) and a phytocompound (hesperetin). It was possible to investigate the cytotoxic effects of phytocompound alone and in combination on the cell line U2OS on several parameters such as colony formation, cellular proliferation, cytotoxicity and genotoxicity. Colony formation was followed by clonogenic assay, cell proliferation was determined by crystal violet (CV) and 3- [4,5-dimethyl-thiazol-2-yl] -2,5-diphenyltetrazolium (MTT) assays. The cytostatic and genotoxic potential was assessed by micronucleus assay with blocked cytokinesis (CBMN) and flow cytometric cell cycle analysis. There were no significant genotoxic effects, however, the hesperetin both isolated and in combination induced cytostatic effects as observed by a decrease in nuclear division index (NDI) and modification of profile phases of the cell cycle. Compared to antineoplastic agents, it was observed that also combinations containing hesperetin did not allow the development of U2OS colonies. Future studies of DNA damage could elucidate the alterations that these compounds cause in this cell line and a comparison with other cell lines would also increase the knowledge on the cytotoxicity and genotoxicity mechanisms.pt
