Abstract: | Este trabalho consistiu em validar dois métodos
cromatográficos que permitissem controlar a concentração de
alguns dos compostos existentes na formulação de um
medicamento com o princípio activo X. Neste sentido foi
validado um método destinado à quantificação do composto Y,
um antioxidante presente na formulação e, um outro método
que permite avaliar o perfil de impurezas na mesma forma
farmacêutica.
Os métodos utilizados foram facultados por um cliente da
Bluepharma e pretendia-se validar estes dois métodos de forma
a aplicá-los posteriormente nas análises de rotina do
medicamento do composto X. Para cada método foram
avaliados os parâmetros pré-definidos para o processo de
validação. Foram realizados testes de estabilidade,
selectividade, precisão do sistema, limite de detecção, limite de
quantificação, linearidade e recuperação, que permitiram
constatar que o método analítico cedido pelo cliente
reproduzirá resultados precisos e fiáveis.
Para o método do conteúdo do composto Y, foi obtida uma
estabilidade de 72 horas para os padrões e apenas 24 horas
para as amostras. Estes tempos garantem que seja feita uma
análise cujas soluções utilizadas não apresentem qualquer
alteração das suas características originais. No estudo da
filtração, também para este método, concluiu-se que o sistema
de filtros PVDF do fornecedor Millipore e Nylon do
fornecedor VWR são os mais adequados comparativamente
com o outro tipo de filtro estudado, uma vez que os resultados
não apresentam uma diferença significativa relativamente à solução não filtrada (p>0,05), com um desvio inferior a 2%.
Em relação à recuperação da substância activa nos níveis de
concentração 60%, 100% e 140%, verificou-se que o método
permite obedecer aos limites de especificação (97%-103%),
cujas recuperações obtidas foram de 100.40%, 100.24% e
100.00%, respectivamente. Esta avaliação permitiu concluir
que o método é exacto.
No método de validação das impurezas do composto X,
também os filtros PVDF e Nylon foram os que menos
interferiram nas soluções filtradas para a generalidade das
impurezas analisadas. A análise da estabilidade da solução
padrão foi de 24 horas, tendo-se obtido um desvio de 0,84%
em relação à injecção feita ao tempo 0 horas. Em relação às
soluções da fórmula reconstituída verificou-se que não se
consegue estabelecer um tempo de estabilidade em relação à
impureza A, pois obteve-se um desvio superior à especificação
após 1,5 horas da primeira injecção. Concluiu-se que de futuro,
as amostras devem ser injectadas logo depois de preparadas. A
partir das análises realizadas para estabelecer os limites de
detecção e quantificação foi possível concluiu-se que o método
permite detectar amostras com concentrações de
1,20x10-5 mg/ml (0.003%), apesar de só conseguir quantificar,
com rigor e precisão, amostras de concentrações iguais ou
superiores a 2,40x10-3 mg/ml (0.006%).
A avaliação de todos os parâmetros permitirão em análises
futuras, no Controlo de Qualidade, identificar e quantificar o
composto Y, bem como as impurezas presentes no
medicamento do composto X.
Neste sentido, o composto Y e as impurezas presentes na
forma farmacêutica são quantificados de forma exacta, precisa,
linear e selectiva (não há influência da substância activa, nem
dos excipientes). This work consisted in the validation of two chromatographic methods that allow control some of the existing compounds in the formulation of a drug with the compound X. So in this sense was validated a method to quantify the Y compound, an antioxidant necessary for the formulation of the drug, and another method to evaluate the impurities expected in the same drug of the compound X. The methods used were provided by a supplied of Bluepharma and sought to validate these two methods in order to apply them later in routine analysis of the drug compound X. For each method were evaluated pre-defined parameters for the validation process. Were performed tests of stability, selectivity, system precision, detection and quantification limits, linearity and recovery, which showed evidence that the analytical method given by the costumer, will provide accurate and reliable results. For the assay method of the compound Y was obtained a stability of 72 hours for the standards and only 24 hours for the samples. By analyzing solutions within these times, it is garanteed that there is no significant change on their behavior. In the filtration study, also for this method, it was concluded that the filter system supplier Millipore PVDF and supplier Millipore PVDF are more appropriate compared to other type of filters studied, since the results do not show a significant difference compared to the unfiltered solution (p>0,05), with a deviation of less than 2%. Regarding to the accuracy test, the analysis of concentration the levels of 60%, 100% and 140%, showed results criterin the specification limits (97%-103%): 100,40%, 100,24% and 100,00% respectively. In the method validation of impurities of compound X, PVDF and Nylon filters showed less interference for the analysis of the impurities evaluated regarding to the other filter brands tested. The stability analysis of standard solution was 24 hours, with a deviation of 0,84% compared to the injection made at time 0 hours. In relation to the solutions of reconstituted formula, it was found that the solutions are not stable regarding to the impurity A, since a deviation greater than the specification was verified 1,5 hours after the first injection. It was concluded that in future, the analysis of tablets, should be injected immediately after preparation. From the analysis carried out to establish the limits of detection and quantification was possible to verify that the method can detect samples with concentrations of 1,20x10-5 mg/ml (0.003%), despite only quantify with accuracy and precision, sample concentrations equal to or greater than 2,40x10-5 mg/ml (0.006%). The evaluation of all parameters will, in future analysis of Quality Control, identify and quantify the compound Y and the known impurities present in drug containing the compound X. In these sense, both the method of content of the compound Y, as the method of impurities in the drug compound X, are quantified without suffering the influence of the active substance or other excipients, presenting the results an acceptable reproducibility and linear. |
