Abstract: | Actualmente ainda não estão estabelecidas as funções exactas da
APP, no entanto vários estudos indicam o seu envolvimento na adesão
celular. Uma vez que a adesão celular, é influenciada pela natureza e
pelo nível de expressão de glicoproteínas existentes na superfície
celular, é importante investigar o efeito da densidade celular na
expressão da APP. Além disso, as variações na densidade celular
podem ser relevantes não só durante o desenvolvimento normal, mas
também em relação a certas patologias, particularmente no que
respeita ao metabolismo anormal de proteínas. Assim, é fundamental
verificar o efeito da densidade celular, não só na expressão da proteína
mas também no seu processamento, em linhas celulares neuronais e
não neuronais.
Os resultados obtidos indicam que a expressão da APP não aumenta
drasticamente com o aumento da densidade celular, mas existe um
pequeno aumento consistente na expressão da APP com a densidade
celular, atingindo o seu valor máximo a aproximadamente 85% de
confluência. Este efeito parece ser menos marcado nas células PC12
do que nas células COS-1. Os níveis mais elevados de expressão da
APP levam a um correspondente aumento na quantidade de APP que é
processada e no sAPP que é secretado para o meio de cultura.
Resultados prévios indicaram que os esteres de forbol estimulam a via
não amiloidogénica, o que resulta numa correspondente diminuição na
produção de Abeta, produzindo um interessante ponto de intervenção
terapêutica. Foi testado o efeito do PMA (ester de forbol) no
processamento da APP, com o aumento da densidade celular.
Observou-se que em ambas as linhas celulares, e a todas as
densidades testadas, o PMA não só estimulou a secreção de sAPP
para o meio, mas também produziu uma diminuição nos níveis
intracelulares de holoAPP. A estimulação do sAPP foi bastante maior
nas células COS-1 quando comparada com as PC12. No entanto, em
ambas as linhas celulares o valor de estimulação estava dependente do
nível basal de sAPP secretado. Estabeleceu-se ainda que a expressão
da APP pode ser regulada por factores presentes no meio extracelular. The precise role of APP remains to be fully elucidated, but various lines of evidence suggest that it may be involved in cell adhesion processes. Thus, since cell adhesion is particularly influenced by the exact nature and level of cell surface glycoproteins, it is important to investigate how increasing cell density (ICD) affects APP expression. Furthermore, because cell density variations may be relevant not only in the course of normal development, but also in relation to specific pathologies, particularly with respect to abnormal protein metabolism, it is important to evaluate the effect of varying cell density not only on APP expression but also on its processing, both for neuronal and non-neuronal cell types. Our results revealed that although the amount of APP expressed did not change dramatically with ICD, there was a small but reproducible increase in APP expression at higher cell confluencies, with the highest level of APP expression being reached when cells were approximately 85% confluent. This effect appeared to be less marked for PC12 cells than for COS-1 cells. Consequently, the higher expression levels of APP led to a corresponding increase in the amount of APP processed and sAPP being secreted into the cell media. Given that previous results indicated that phorbol esters stimulate the non-amyloidogenic pathway at the expense of reducing Abeta production, thus providing an interesting therapeutic focus, we tested the effect of the phorbol ester PMA on APP processing at ICD. We observed that in both PC12 and COS-1 cells, and at all densities tested, PMA not only stimulated sAPP secretion into the medium, but also produced a corresponding decrease in the intracellular levels of holoAPP. The stimulation of sAPP secreted was significantly higher in COS-1 cells compared to PC12 cells. However, in both cell types the fold increase was dependent on the basal level of sAPP secreted. Further experimentation went on to elucidate that the increase in APP expression can be regulated by factors present in the extracellular milieu. |
